Non-competitive antagonism occurs when an antagonist reduces the effect of an agonist in a way that cannot be overcome by increasing the concentration of the agonist, often by binding to an allosteric site or irreversibly to the receptor. This results in a decrease in the maximum response achievable by the agonist, altering the efficacy but not the potency of the agonist-receptor interaction.
